Zeiss Lattice Light-Sheet microscope

Applications

Lattice light-sheet (LLS) microscopy technology was developed by the laboratory of Nobel Prize winner Eric Betzig and was the subject of a very first publication in the journal Science in 2014. This technology allows 4D imaging (also called volumetric imaging), i.e. to observe living samples with a high spatial resolution (allowing to visualize subcellular structures) combined with a very high temporal resolution, with low phototoxicity and low photobleaching. While the first versions of LLSM were very heavy to use, Zeiss made a very user-friendly version of this technology. Zeiss Lattice Lightsheet 7 allows gentle volumetric imaging at high speed (e.g. 1 cell volume acquired in less that 1 second, depending on desired resolution and number of channels) and near-isotropic spatial resolution (approx. 350-400 nm). Of note, samples must be of reasonable thickness (< 200 µm).

Note: Volumetric high-resolution imaging produces large amounts of data (Gb to Tb!) in a very short amount of time. Projects must be discussed with platform specialists regarding data storage and data analysis. Please, contact us.

Technical description

  • Zeiss Lattice Lightsheet 7, inverted configuration, fully motorized stage
  • Excitation laser lines (diodes): 488 nm (10 mW), 561 nm (10 mW) and 640 nm (5 mW)
  • Detectors: None
  • Objectives: special optics developed by Zeiss specifically for lattice light-sheet, water auto-immersion system for long-term imaging. Note: stage and optics allow imaging in any high quality (#1.5) glass-bottom cell culture dishes (chambers such as Ibidi µ-slide or LabTek, 35 mm diameter Petri dishes, 24 or 96-well dishes…).
  • Drift correction: fully controlled stage
  • UV-Vis light source: None
  • Camera: PCO.edge 4.2 sCMOS camera (QE 82%, pixel size 6.5 µm x 6.5 µm, resolution 2048 x 2048)
  • Incubator: integrated (control of CO2, temperature and humidity; possibility to add N2 supply to work in hypoxic conditions)
  • Antivibration table (compressed air)
